What is RBCs( red blood cells) ?
Red blood cell-Red blood cells are the types of blood cell its scientific name is erythrocytes this is the most abundent cell in the blood. 4.5-6.5 million per macrolitres of blood cells are present in human body.
Its form in the bone marrow and the mature RBC forms do not have a nucleus. The  RBC shaped is Bioconcave disk. The lifespan of RBC is about 90 -120 days.Its about 6-8 micrometres in diameter.
Hemoglobin(iron-protein)  is found in the RBC. Haemoglobin carries oxygen from the lungs to the the rest of the body and carbon dioxide binds to the RBC and taken  to the lungs to be Exhaled.

What is RBC count?
Red blood cell count is blood test that you find out how many red blood cells (RBCs) you have.It’s also known as erythrocytes count this test is important because RBCs contain hemoglobin which carry oxygen to to your body’s tissue.The number of RBCs you have can affect how much oxygen your tissue receive, your tissue need oxygen to function.
Requirement
  1. Neubaur chamber 
  2. Whole blood(EDTA blood) 
  3. Hayem’s fluid 
  4. Cover slip
  5. Microscope 

Procedure of RBC count-

  • First we take a clean and dry test tube and 
  • Take 3980 microliter of hyme’s fluid and add 20 microlitre of blood and (for dilution preparation). 
  • Mix well and wait for 2 to 3 minutes 
  • After that we take counting chamber  and charge the chamber with dilution (10 microliter) and cover with cover slip. 
  • And wait for 1-2 minutes and examine under microscope (low power then 40x).
  •  After that calculate and give the result ( total number of RBC x 10,000).
The calculation of RBC count-
Calculation
= number of RBC count X dilution / Depth X  area count
= No.  of total RBCs count X 10,000

 lowRBC count-
  • Fatigue 
  • Shortness of breathing 
  • Dizziness
  • Weakness 
  • Increased heart rate 
  • Headache 
  • Pale skin 
High RBC count –
  • Joint pain 
  • Shortness of breathing
  • Itching skin, particularly after a shower or bath 
  • Sleep disturbance

What is the composition of RBC diluting fluid?(Hyme’s fluid ).

This is an isotonic field which consists of the following constituents(in g/100ml)
  • Sodium chloride – 0.5 ml
  • Sodium sulphate – 2.5 ml
  • Mercuric chloride – 0.25 ml
  • Distilled water – 100 ml
             The sodium sulphate discourage  clumping of the erythrocytes and the mercuric chloride preservative.
